$IOVA: The Company Said “Positive Data.” The Stock Dropped 25%. Both Are Right

Iovance's lung cancer TIL data showed a 59% response rate in a biomarker subgroup and 11% outside it, and the stock gave back a quarter of its value premarket. The subgroup math, the real balance sheet, what November 5 has to show, and scenario zones for the week ahead.

$IOVA: The Company Said “Positive Data.” The Stock Dropped 25%. Both Are Right

On Wednesday evening, Iovance Biotherapeutics (Nasdaq: IOVA) told the market it would host a webcast to discuss "positive data" for its TIL cell therapy in metastatic non-small cell lung cancer. On Thursday morning, the data landed, and the stock lost roughly a quarter of its value before the opening bell, trading near $11 in premarket against Wednesday's $14.81 close. Both the company and the market are telling the truth. That is what makes this one interesting.

The press release is genuinely positive if you read it the way Iovance wants you to. In the Phase 2 IOV-LUN-202 trial, 17 of 69 patients with previously treated non-squamous metastatic NSCLC responded to a single infusion of lifileucel, an objective response rate of 24.6%, consistent with the 25.6% reported earlier in a smaller cohort. In a biomarker-defined subgroup, the response rate was 59.1%, including both of the study's complete responses. The company announced a new trial, POSITILITY, enrolling from November with registrational intent, plus a frontline study, OPTILISM, planned for 2027, where an early seven-patient cohort combining lifileucel with pembrolizumab produced six responses.

The market read the same document differently, and its reading is also defensible. The 59.1% number comes from a retrospective slice: patients who are KRAS wild-type, HER2 wild-type, and current or former smokers. That subgroup was 22 of 69 patients, under a third of the study, but produced 13 of the 17 responses. Flip that around and the arithmetic gets uncomfortable. In patients with KRAS-mutant tumors, roughly half the enrolled and assessed population, the response rate was 11.4%. The therapy that was supposed to address a market Iovance itself has described as about seven times the size of its approved melanoma indication now looks, on this dataset, like a therapy for a biomarker-enriched segment the company estimates at roughly 45% of non-squamous metastatic NSCLC cases. The broad-label lung story did not die on Thursday, but it got materially smaller and moved further away, and the stock had been priced for the big version.

Context matters here, because this is not a broken biotech bleeding out. Iovance closed Wednesday up roughly 650% over twelve months. Amtagvi, the melanoma version of the same therapy, is selling: record revenue of $99.3 million in the second quarter, and a full-year guidance raise on September 29 to $410 to $420 million, implying nearly 60% annual growth. The company says its cash funds operations into the second half of 2028. Third-quarter earnings land Thursday, November 5, one week from today. So the question the premarket is answering with panic is narrower than it looks: not "is Iovance viable," but "what is the lung program worth now, and how much of the last three months' run was riding on it?"
